A laranja Pêra (Citrus sinensis) e o limão Taiti (Citrus latifólia), estão entre as frutas cítricas mais amplamente cultivadas e consumidas, sendo o Brasil um dos principais produtores e exportadores mundiais. Essas frutas são ricas em compostos bioativos, incluindo vitamina C e fenólicos. O presente estudo teve como objetivo investigar a bioacessibilidade e o potencial bioativo dos polifenóis da laranja e do limão, avaliando suas polpas e cascas. Determinou-se a bioacessibilidade e a bioatividade das frações bioacessíveis em células humanas Caco-2 após a simulação gastrointestinal, e os principais compostos fenólicos foram identificados por CLAE-DAD. No Capítulo 2, casca de laranja apresentou valores de 492,78 no extrato hidroetanólico a 167,04 µmol TEAC/100g na fase intestinal para atividade antioxidante (AA) e de 848,17 no extrato hidroetanólico a 443,11 mg GAE/100g na fase intestinal para capacidade redutora (CRF), com índice de bioacessibilidade (IB%) de 33,90 para AA e 52,24 para CRF. A polpa variou de 27,26 nas amostras in natura a 42,25 µmol TEAC/100g na fase intestinal para AA e de 104,89 nas amostras in natura a 150,83 mg GAE/100g na fase intestinal para CRF, com IB% de 155,02 para AA e 69,54 para CRF. O extrato da polpa e suas frações bioacessíveis, quando avaliados em células Caco-2 em concentrações de até 20 e 60 µmol, respectivamente, não apresentaram citotoxicidade, com viabilidade celular superior a 50%. Hesperidina e naringina foram identificados como fenólicos majoritários nas amostras de extrato de laranja e na fase intestinal. No Capítulo 3, para CRF e a AA a casca de limão apresentou valores de 1768,17 no extrato hidroetanólico a 760,00 mg GAE/100g na fase intestinal para CRF e de 644,45 no extrato hidroetanólico a 173,73 µmol TEAC/100g na fase intestinal para AA, com IB% de 42,98 para CRF e 26,96 para AA. A polpa variou de 156,27 nas amostras in natura a 93,78 mg GAE/100g na fase intestinal para CRF e 27,98 nas amostras in natura a 82,79 µmol TEAC/100g na fase intestinal para AA, com IB% de 60,01 para CRF e 259,88 para AA. A avaliação da citotoxicidade em células Caco-2 demonstrou que o extrato da polpa e suas frações não exibiram citotoxicidade celular em concentrações de até 20 e 60 µmol, respectivamente. A hesperidina foi o principal fenólico, seguida pelo ácido clorogênico. No Capítulo 4, a codigestão das cascas de laranja e limão variou de 569,26 no extrato hidroetanólico a 165,04 µmol TEAC/100g na fase intestinal para AA e de 1434,83 no extrato hidroetanólico a 538,67 mg GAE/100g na fase intestinal para CRF, com IB% de 28,99 para AA e 37,54 para CRF. Para a polpa, variou de 27,40 nas amostras in natura a 71,91 µmol TEAC/100g para AA e de 150,81 nas amostras in natura a 135,11 mg GAE/100g na fase intestinal para CRF, com IB% de 262,49 para AA e 89,59 para CRF. A partir das diferenças e os percentuais de sinergia, os valores obtidos foram negativos, indicando ausência de efeitos sinérgicos na codigestão das amostras. De modo geral, houve variações da CRF e da AA ao longo da digestão das amostras, porém uma fração considerável de fenólicos da polpa e da casca da laranja e do limão são bioacessíveis. Embora as cascas apresentem maior concentração de fenólicos, a fração efetivamente disponível para absorção foi mais expressiva nas polpas. Os resultados demonstram o potencial bioativo da laranja e do limão. Contudo, fazem-se necessários estudos contínuos e complementares para esclarecer as interações entre frutas cítricas, além de futuros estudos para investigar a bioatividade e efeitos à saúde de polifenóis de frutas cítricas. Abstract: Pêra orange (Citrus sinensis) and Taiti lemon (Citrus latifólia) are among the most widely cultivated and consumed citrus fruits, and Brazil is one of the world?s leading producers and exporters. These fruits are rich in bioactive compounds, such as vitamin C and phenolics. The present study aimed to investigate the bioaccessibility and bioactive potential of polyphenols from orange and lemon, evaluating their pulp and peel. The bioaccessibility and bioactivity of the bioaccessible fractions were determined in human Caco-2 cells after gastrointestinal simulation, and the main phenolic compounds were identified by HPLC-DAD. In Chapter 2, the peel the orange showed values ranging from 492.78 in the hydroethanolic extract to 167.04 µmol TEAC/100g in the intestinal phase for antioxidant activity (AA), and from 848.17 in the hydroethanolic extract to 443.11 mg GAE/100g in the intestinal phase for Folin-Ciocalteu reducing capacity (FRC), with a bioaccessibility index (BI%) of 33.90 for AA and 52.24 for RFC. The pulp ranged from 27.26 in natura samples to 42.25 µmol TEAC/100g in the intestinal phase for AA, and from 104.89 in natura samples to 150.83 mg GAE/100g in the intestinal phase for RFC, with BI% of 155.02 for AA and 69.54 for FRC. The pulp extract and its bioaccessible fractions, when evaluated in Caco-2 cells at concentrations lower than 20 and 60 µmol, respectively, showed no cytotoxicity, with cell viability higher than 50%. Hesperidin and naringin were identified as the major phenolics in orange extract samples and in the intestinal phase. In Chapter 3, the peel the lemon showed values ranging from 1768.17 in the hydroethanolic extract to 760.00 mg GAE/100g in the intestinal phase for FRC, and from 644.45 in the hydroethanolic extract to 173.73 µmol TEAC/100g in the intestinal phase for AA, with BI% of 42.98 for FRC and 26.96 for AA. The pulp ranged from 156.27 in natura samples to 93.78 mg GAE/100g in the intestinal phase for FRC, and from 27.98 in natura samples to 82.79 µmol TEAC/100g in the intestinal phase for AA, with BI% of 60.01 for FRC and 259.88 for AA. Cytotoxicity evaluation in Caco-2 cells demonstrated that the pulp extract and its fractions showed no cytotoxicity at concentrations lower than 20 and 60 µmol, respectively. Hesperidin was the main phenolic compound, followed by chlorogenic acid in both the lemon extract and the intestinal phase. In Chapter 4, codigestion of orange and lemon peel showed values ranging from 569.26 in the hydroethanolic extract to 165.04 µmol TEAC/100g in the intestinal phase for AA, and from 1434.83 in the hydroethanolic extract to 538.67 mg GAE/100g in the intestinal phase for FRC, with BI% of 28.99 for AA and 37.54 for FRC. The pulp ranged from 27.40 in natura samples to 71.91 µmol TEAC/100g in the intestinal phase for AA, and from 150.81 in natura samples to 135.11 mg GAE/100g in the intestinal phase for FRC, with BI% of 262.49 for AA and 89.59 for FRC. Based on the differences and synergy percentages, the obtained values were negative, indicating the absence of synergistic effects during codigestion. Overall, variations in FRC and AA were observed throughout the digestion of the samples, however, a considerable fraction of phenolic compounds from both the pulp and peel of orange and lemon are bioaccessible. Although the peels showed higher phenolic concentrations, the fraction effectively available for absorption was more expressive in the pulps. These results demonstrate the bioactive potential of orange and lemon. However, further and complementary studies are needed to clarify interactions between citrus fruits and to investigate the bioactivity and health effects of citrus polyphenols. |
