Title: Trichoderma koningiopsis fermentation in Airlift bioreactor for bioinputs production
Author: Camargo, Aline Frumi
Abstract: The traditional food production model faces difficulties due to the excessive use of agrochemicals, which result in environmental contamination and impacts health and agricultural productivity. Bioherbicides, which are biological products for controlling invasive plants, have emerged as an alternative to reduce the use of agrochemicals. In Brazil, the bioinput market has expanded, with new legislation that encouraging microbiological products for pest control. To this end, the use of bioreactors and optimized fermentation processes are crucial to ensure the efficiency and quality of these products. Therefore, the main objective of this thesis was to optimize a fermentation process using microalgal biomass in an airlift bioreactor, evaluate its bioherbicidal potential, and explore additional biotechnological applications of the resulting bioproduct. To this end, we studied the parameters of the fermentation process of a fungal extract and quantified enzymes involved in the bioherbicidal activity (amylase, cellulase, laccase, lipase, and peroxidase) in the extracts obtained. We evaluated agronomic efficiency through growth promotion tests and soybean nodulation, soybean seed germination and in vitro control of phytopathogens. We dedicated ourselves to investigating the cytotoxicity of the fermented and optimized fungal extract, which was fractionated using solvents of different polarities to evaluate in vitro antitumor activities in A549 tumor cell lines and non-tumor Vero cells. Finally, we studied the behavior of the same fermentation process, but using a different substrate, microalgae, cultivated as a sewage treatment from sanitary effluents. The results highlight an optimized fermentation condition, with agronomic and industrial interest in the production of enzymes. The optimized condition has the following characteristics: 200 g of wet microalgal biomass, an aeration of 8 LPM, temperature of 28 ºC, pH 8.3, 400 mL of antifoam diluted in the fermentation medium and final volume of 2.0 L. The fermented extract proved efficient in the biological control of pathogens and in promoting soybean growth. In addition, the extract presented low cytotoxicity because it did not affect the proliferation of normal cells. Adapting the process to use residual microalgae also showed feasibility, making bioprocess more sustainable and economical. The research contributes to the development of more sustainable agricultural practices, underscoring the importance of bioherbicides and the use of efficient bioprocesses to produce bioinputs, as allowed us to expand our understanding of fermentations in airlift bioreactors, highlighting the importance of optimizing bioprocess conditions to improve the productivity and quality of bioproducts.
