dc.description.abstract O mofo-branco, causado por Sclerotinia sclerotiorum, é uma doença com grande potencial de reduzir a produtividade de diversas culturas de interesse econômico como a soja, algodão e feijão. O uso de fungicidas no controle da doença, além de causar grandes impactos negativos ao meio ambiente e à saúde das pessoas, também não é totalmente eficiente no controle das estruturas de resistência do fitopatógeno, os escleródios. Diante disso, o controle biólogo pode ser visto como uma ferramenta no controle da doença, permitindo a redução do inóculo do fitopatógeno com maior eficiência e especificidade e reduzindo os riscos de contaminação ambiental. Atualmente, poucas espécies do gênero Trichoderma são comercializadas, portanto, é importante o isolamento e a seleção de novos bioagentes para o desenvolvimento e disponibilidade de novos produtos e formulações no mercado, permitindo o aumento da oferta e estimulo da geração de demanda. Dessa forma, o objetivo do trabalho foi avaliar in vitro o potencial de isolados de Trichoderma spp., na inibição micelial e na redução da viabilidade de escleródios de Sclerotinia sclerotiorum. Foram analisados 8 isolados coletados de solos do município de Brunópolis/SC (1A; 36A; 98A; 99A; 111B; 112B; 36B; 102) e um isolado do produto comercial Thichodermil. Foi realizado o teste de pareamento em placas de Petri em duas metodologias, na primeira metodologia (a), foi inoculado um disco de micélio do fitopatógeno, centralizado no meio da placa de Petri contendo BDA (Batata-Dextrose-Ágar), e dois discos de micélio do antagonista, um em cada extremidade da placa. Na metodologia (b), um disco de micélio do fitopatógeno foi inoculado em uma das extremidades do meio de cultura e um disco de micélio do antagonista foi inoculado na outra extremidade. Com os dados, foram calculados a porcentagem de inibição e porcentagem de redução da velocidade do crescimento do fitopatógeno. Para a avaliação da redução da viabilidade de escleródios do fitopatógeno, seis escleródios produzidos em BDA com 25 dias de idade foram transferidos para placas contendo colônias de Trichoderma e mantidas incubadas por 15 e 40 dias. A viabilidade dos escleródios foi avaliada pelo teste de Trifenil Cloreto de Tetrazólio (0,5%), em que foi considerado viável os escleródios de coloração rosada e não viável os escleródios de coloração amarelada. Para ambos os testes foi utilizado o delineamento inteiramente casualizado, com 10 tratamentos e cinco repetições. Os dados coletados foram submetidos à análise de variância no software R e comparadas pelo teste de Scott Knott, ao nível de significância de 5%. Os resultados demonstraram que todos os isolados de Trichoderma mostram potencial antagônico sobre S. sclerotiorum, todavia, os isolados com maior eficiência do controle micelial são o 1A, 112B, 36B e 99A. Quanto a redução da viabilidade dos escleródios, com 15 dias nenhum isolado demonstrou capacidade de controle da estrutura, todavia, com 40 dias de incubação, os isolados 1A, 102B e TCOM demonstraram serem capazes de reduzir a viabilidade dos escleródios. Notou-se o grande potencial desses isolados nativos da região de Santa Catarina no controle do mofo-branco. pt_BR
dc.description.abstract White mold, caused by Sclerotinia sclerotiorum, is a disease with great potential to reduce the productivity of several crops of economic interest such as soybeans, cotton and beans. The use of fungicides to control the disease, in addition to causing great negative impacts to the environment and people's health, is also not fully efficient in controlling the resistance structures of the phytopathogen, the sclerotia. Therefore, biological control can be seen as a tool in the control of the disease, allowing the reduction of the phytopathogen inoculum with greater efficiency and specificity, reducing the risks of environmental contamination. Currently, few species of the Trichoderma genus are commercialized, therefore, it is important the isolation and selection of new bioagents for the development and availability of new products and formulations on the market, allowing an increase in supply and stimulation of demand generation. Thus, the objective of this work was to evaluate in vitro the potential of Trichoderma spp. isolates in mycelial inhibition and in reducing the viability of Sclerotinia sclerotiorum sclerotia. Eight isolates collected from soils in the municipality of Brunópolis (1A; 36A; 98A; 99A; 111B; 112B; 36B; 102) and 1 isolate from the commercial product Thichodermil were analyzed. The pairing test in Petri dishes was carried out in two methodologies, in the first methodology (a), a disc of phytopathogen mycelium was inoculated, centered in the middle of the Petri dish containing PDA (Potato-Dextrose-Agar), and two discs of antagonist mycelium, one at each end of the plate. In methodology (b), a mycelium disc of the phytopathogen was inoculated at one end of the culture medium and an antagonist mycelium disc was inoculated at the other end. With the data, the percentage of inhibition and percentage of reduction in the growth velocity of the phytopathogen were calculated. For the evaluation of the reduction of the sclerotia viability of the phytopathogen, 6 sclerotia produced in PDA with 25 days of age were transferred to plates containing colonies of Trichoderma and kept incubated for 15 and 40 days. The sclerotia viability was evaluated by the Triphenyl Tetrazolium Chloride (0.5%) test, in which the pink colored sclerotia were considered viable and the yellowish colored sclerotia were not viable. For both tests, a completely randomized design was used, with 10 treatments and five replications. The collected data were submitted to analysis of variance in the R software and compared by the Scott Knott test, at a significance level of 5%. The results showed that all Trichoderma isolates show antagonistic potential against S. sclerotiorum, however, the isolates with the highest efficiency of mycelial control are 1A, 112B, 36B and 99A. Regarding the reduction of sclerotia viability, at 15 days no isolate showed the ability to control the structure, however, at 40 days of incubation, isolates 1A, 102B and TCOM were able to reduce sclerotia viability. It was noted the great potential of these isolates native to the region in the control of white mold. en
