dc.description.abstract O Alternaria porri é um fungo altamente destrutivo para a cultura da cebola, entretanto a transmissão via semente ainda possui relevância indefinida. O objetivo deste trabalho foi avaliar a influência do fungo A. porri no desenvolvimento de sementes de cebola (Allium cepa L.) e sua capacidade de contaminar plântulas através da infecção via semente. O fungo foi obtido através da micoteca do laboratório de Fitopatologia da Universidade Federal de Santa Catarina, isolado e repicado em placas de Petri contendo meio BDA. As sementes de cebola foram acondicionadas sobre as colônias fúngicas promovendo o contato direto com o micélio do fungo assegurando a infecção das mesmas, assim que alguma das sementes apresentou protrusão radicular estas foram retiradas e secas em papel filtro. Em seguida foram submetidas aos testes de germinação, emergência e sanidade. Para a avaliação da germinação, as sementes foram dispostas em caixas do tipo gerbox com papel germitest umedecido e incubado a 25ºC e fotoperíodo de 12 h, avaliando-se o número de sementes germinadas aos seis e 12 dias,o delineamento foi inteiramente casualisado com quatro repetições. O teste de sanidade foi realizado pelo método de plaqueamento em meio Ágar sólido (BDA), na qual as sementes foram dispostas em placas de Petri contendo meio BDA e incubadas em câmara de crescimento, as avaliações foram realizadas aos sete e 14 dias quanto à presença de colônias fúngicas na semente. Para o teste de emergência as sementes foram semeadas em caixas do tipo gerbox contendo areia autoclavada, umedecidas e incubadas. As avaliações foram realizadas aos sete e 21 dias contabilizando a quantidade de plântulas normais emergidas e realizando a classificação de plântulas aos 21 dias, o delineamento foi inteiramente casualisado com seis repetições. O fungo A. porri causou redução significativa na germinação das sementes de cebola aos 14 dias, tendo sua contaminação comprovada pelo teste de sanidade apresentando alta porcentagem de infecção nas sementes. O teste de emergência apresentou diferença significativa aos 21 dias, reduzindo significativamente o número de plântulas normais emergidas. A classificação de plântulas diferiu para plântulas normais, que foi maior na testemunha, e para plântulas anormais superior no tratamento com sementes infectadas, afirmando a capacidade do fungo em contaminar plântulas a partir da transmissão via semente, podendo contribuir para disseminação da doença no campo. pt_BR
dc.description.abstract The Alternaria porri is a highly destructive fungus for the onion culture, however the transmission via seed still has indefinite relevance. The objective of this work was to evaluate the influence of A. porri fungus on the development of onion seeds (Allium cepa L.) and its ability to contaminate plants through seed infection. The fungus was obtained from the mycological specimen collection of the University of Santa Catarina’s Phytopathology Laboratory , isolated and repicted to Petri dishes containing PDA environment. The onion seeds were laid on the fungal colonies promoting the direct contact with the mycelium of the fungus ensuring their infection, as soon as the first seed presented root protrusion they were removed and dried on filter paper. Soon afterwards, they were submitted to germination, emergency and health tests. For the germination evaluation, the seeds were arranged in gerbox boxes with germitest paper moistened and incubated at 25ºC and photoperiod of 12 h, evaluating the number of seeds germinated on the 6th and 12th days, the line ation was entirely randomized with 4 repetitions. The health test was carried out using the Agar solid plating method (PDA), which the seeds were arranged in Petri dishes containing PDA environment and incubated in a growth chamber, the evaluations were performed on the 7th and 14th days to check the presence of fungal colonies in the seed. For the emergency test, the seeds were seeded in gerbox boxes containing autoclaved sand, moistened and incubated. The evaluations were carried out on the 7th and 21th days counting the amount of normal seedlings emerged and performing seedling classification at the 21st day, the lineation was completely randomized with 6 repetitions. The A. porri fungus caused a significant reduction in germination of onion seeds on the 14th day, and its contamination was confirmed by the health test, presenting a high percentage of infection in the seeds. The emergency test showed a significant difference on the 21th day, significantly reducing the number of normal emerged seedlings. The classification of seedlings differed of the normal seedlings, which was higher in the control, and for abnormal seedlings higher in the treatment with infected seeds, affirming the ability of the fungus to contaminate seedlings from the transmission via seed. en
