O presente trabalho investiga a extração e purificação de isoflavonas a partir de cotilédones de soja (Glycine max L.), visando a obtenção de extratos bioativos para aplicação em medicina regenerativa. O estudo foca na conversão de isoflavonas glicosiladas em suas formas agliconas (daidzeína e genisteína) por meio de hidrólise ácida, buscando aumentar a biodisponibilidade e eficácia para a regeneração tecidual em comparação às terapias convencionais. Foi utilizada extração assistida por ultrassom com soluções hidroalcoólicas de metanol e etanol. A caracterização fitoquímica por cromatografia líquida de alta eficiência (HPLC-MS) confirmou a presença dos compostos de interesse, identificando a daidzina no extrato metanólico purificado e a daidzeína no extrato etanólico hidrolisado como as maiores concentrações obtidas, sendo 62,25 mg/g e 28,95 mg/g, respectivamente. Nos ensaios de atividade antioxidante pelo método de eliminação de radicais livres ABTS, o extrato etanólico bruto apresentou o melhor desempenho EC50 próximo a 3,5 mg/mL, sugerindo uma interação entre as diferentes substâncias, potencializando a capacidade antioxidante da matriz vegetal. Adicionalmente, o extrato etanólico purificado hidrolisado demonstrou potencial antibacteriano contra Staphylococcus epidermidis, com cerca de 62,14% de inibição, em faixa equivalente ao antibiótico Ampicilina, com 62,55% de inibição. Os ensaios de viabilidade celular in vitro com fibroblastos L929 indicaram biocompatibilidade em concentrações de até 500 µg/mL. Em doses de 100 µg/mL, as amostras demonstraram efeito bioestimulador, com destaque para a daidzeína e o extrato metanólico bruto após 72 horas de exposição. Os resultados concluem que as isoflavonas de soja possuem potencial terapêutico seguro e eficaz, atuando como agentes antioxidantes e proliferativos com aplicação promissora na engenharia de tecidos. Abstract: This work investigates the extraction and purification of isoflavones from soybean cotyledons (Glycine max L.), aiming to obtain bioactive extracts for application in regenerative medicine. The study focuses on the conversion of glycosylated isoflavones into their aglycone forms (daidzein and genistein) through acid hydrolysis, seeking to increase bioavailability and efficacy for tissue regeneration compared to conventional therapies. Ultrasound-assisted extraction with hydroalcoholic solutions of methanol and ethanol was used. Phytochemical characterization by high-performance liquid chromatography (HPLC-MS) confirmed the presence of the compounds of interest, identifying daidzin in the purified methanolic extract and daidzein in the hydrolyzed ethanolic extract as the highest concentrations obtained, being 62.25 mg/g and 28.95 mg/g, respectively. In antioxidant activity assays using the ABTS free radical scavenging method, the crude ethanolic extract showed the best EC50 performance near 3.5 mg/mL, suggesting an interaction between the different substances, enhancing the antioxidant capacity of the plant matrix. Additionally, the purified hydrolyzed ethanolic extract demonstrated antibacterial potential against Staphylococcus epidermidis, with approximately 62.14% inhibition, in a range equivalent to the antibiotic Ampicillin, with 62.55% inhibition. In vitro cell viability assays with L929 fibroblasts indicated biocompatibility at concentrations up to 500 µg/mL. At doses of 100 µg/mL, the samples demonstrated a biostimulatory effect, with daidzein and the crude methanolic extract standing out after 72 hours of exposure. The results conclude that soy isoflavones possess safe and effective therapeutic potential, acting as antioxidant and proliferative agents with promising applications in tissue engineering. |
