dc.description.abstract O pré-condicionamento cerebral se refere a um estado de tolerância transitória do tecido nervoso a um estimulo letal evocado por um insulto moderado prévio. O pré-condicionamento pode ser evocado por estímulos físicos ou químicos, como o pré-condicionamento com N-metil-D-aspartato (NMDA). O pré-condicionamento com NMDA pode ser promovido in vivo pela administração de uma dose sub-convulsivante de NMDA, com efeito neuroprotetor contra a toxicidade do cainato ou isquemia, além de prevenir convulsões e morte neuronal induzida pelo ácido quinolínico (QA) em camundongos. Este trabalho teve como objetivo principal avaliar a participação das vias de sinalização intracelulares da Proteína Cinase C (PKC), da Proteína Cinase dependente de AMP cíclico (PKA), da Proteína Cinase regulada por sinais extracelulares (ERK), e da Proteína Cinase dependente de Ca2+ /calmodulina II (CaMKII) no fenômeno da neuroproteção atingida por meio de um pré-condicionamento químico. Os camundongos foram pré-condicionados com NMDA (75mg/kg, i.p) 24 h antes da infusão de AQ (4 μl, 36,8nmol i.c.v.) e receberam inibidores específicos das vias de sinalização i.c.v 15 minutos antes do pré-condicionamento, ou 15 minutos antes da infusão de QA. A inibição da via de sinalização da PKA reverteu a proteção evocada por NMDA, e a inibição da via da ERK reverteu parcialmente esta proteção, reduzindo a taxa de proteção a 12,5%. O tratamento com os inibidores da PKC e da CaMKII não alteraram a taxa de proteção.A inibição da via de sinalização da PKC causou uma taxa de morte elevada quando da injeção de NMDA seguida da infusão de QA e quando seguida somente da infusão de QA. A inibição da via da ERK causou uma mortalidade elevada quando seguida da infusão de QA. A inibição das vias de sinalização não alterou a latência e duração das convulsões causadas pelo QA. Os resultados aqui obtidos sugerem que as vias de sinalização da PKA e da ERK exerçam um papel crucial na obtenção de um estado neuroprotetor induzido pelo pré-condicionamento por NMDA. pt_BR
dc.description.abstract Brain preconditioning refers to a state of transient tolerance evoked by a brief insult in which the nervous tissue becomes more resistant to a lethal posterior insult. This preconditioning may be evoked by physical or chemical stimuli, such as the preconditioning by N-methyl-D-aspartate (NMDA). The preconditioning by NMDA may be promoted in vivo by the administration of a sub-convulsing NMDA dose, with a neuroprotective effect against kainate toxicity or ischemia, and it can prevent seizures and neuronal death induced by the infusion of Quinolinic Acid (QA) in mice. This study aims, as its primary objective, to evaluate the participation of the Protein Kinase C (PKC), cAMP-dependent Protein Kinase (PKA), Extracellular signal regulated Kinase (ERK), Ca2+ /calmodulin II dependent Protein Kinase (CaMKII) signaling pathways in the neuroprotection phenomenon attained through chemical preconditioning. Adult Swiss Male mice were preconditioned with NMDA (75mg/kg, i.p) 24 h prior to the infusion of AQ (4 μl, 36,8nmol i.c.v.) and were treated with specific inhibitors of the aforementioned signaling pathways either 15 minutes before the preconditioning, or 15 minutes before the infusion of QA. The inhibition of the PKA pathway abolished the protection evoked by NMDA, and the inhibition of ERK pathway significantly diminished this protection, reducing the protection rate to 12.5%. Treatment with the PKC and CaMKII inhibitors did not alter the protection rate. The inhibition of the PKC pathway resulted in a high mortality rate when carried out before NMDA preconditioning followed by QA infusion, and when carried out before the QA infusion. The inhibition of the ERK pathway resulted in a high mortality rate when followed by the infusion of QA. The inhibition of signaling pathways in this model did not alter the latency or duration of the QA-induced seizures. The results herein presented suggest that the PKA and ERK pathways have a crucial role in the achievement of a neuroprotective state induced by the NMDA preconditioning. pt_BR
